"Knock-in" mice are also known as transgenic mice. Genes responsible for particular traits or disease susceptibility are chosen and extracted. Next they are injected into fertilized mouse eggs. Embryos are implanted in the uterus of a surrogate mother. The selected genes will be expressed by some of the offspring.

Since the first gene transfers into mice were successfully executed in 1980, transgenic mice have allowed researchers to observe experimentally what happens to an entire organism during the progression of a disease. Transgenic mice have become models for studying human diseases and their treatments.

Gene knock-in. The opposite which is more commonly used is knock-out which is why you would commonly here knockout mice. What this means is the a particular gene was selectively knock out to study the effect of having this gene defect making it a good animal model for us to study using. This and knock in are both becoming great tools to use in functional genetics. Gene knock in would be the direct opposite to knock out. A foreign gene could have been introduced to the organism making it transgenic. This is how we create genetically modified food and we do pharming(making transgenic sheep make proteins like insulin). Also knock in could help us see the effect of having a gene over-expressed or a mutation by insertion like in cancer; making it a good model to study the role of the particular gene in out body
As some people have already said, it's the opposite to Knock-out. It allows you to insert a new gene in the site of an old one. For example, I could place a gene which is normally switched on in the liver in place of one which is switched on in the skin. Then I could see what effect that having this gene in place has. So you're removing a gene, and replacing it with a new one. Also, it is slightly different to transgenics, where you are just adding a gene, not replacing.
