Binding equilibrium is a way to determine the characteristics of a compound (ligand) when it binds to a receptor (protein, DNA. etc). At equilibrium only a certain percentage of binding sites will be occupied. Here are a few equations.

A binding equilbrium is determined by the following equation:

[A] + [R] <----> [AR]

where,

[A] = concentration of ligand
[R] = concentration of receptor
[AR] = concentration of ligand-receptor complex

The rate for the forward reaction is k(1) and for the reverse reaction k(-1)

The affinity constant (Ka) is given as k(1)/k(-1)

The fractional occupancy of a set of receptors at a given concentration of ligand at equilibrium is given as:

[AR]/Rt = ([A]Ka) / (1 + [A]Ka)

These constants are often highly variable, as illustrated in the present paper by two examples, binding of salicylate to human serum albumin and of oxygen to hemoglobin. In order to avoid over-interpretation of binding constants it is pointed out that the best-fit solution, obtained by least-squares fitting, may be supplemented by several, e.g. thirty, acceptable solutions. It is further shown how the 30 sets of acceptable binding constants, plotted as Klotz' affinity profiles, can serve for evaluation of cooperative effects.
