Believe it or not, on the bus. You could fly but it'll cost you, and you'll miss out on some great countryside. The only time I ever flew after arriving in south America was when the Argentinian Peso had crashed (2002), making a trip from northern Argentina down to Tierra del Fuego cheaper than the ($30) bus.

Flying will be the most comfortable and shortest journey but is also the most expensive. LAN Peru http://www.lan.com/index.html is the airline you want. Catching the bus is the cheapest option but is also the longest in journey - I reckon you're looking at 20+ hours. Can't remember the name of the bus company but you should be able to google it.
