What or where is the best way to spend a day and a night in the UK?

Everybody goes to london at the end of the day, why not come see the capital of our fine county!
I would recommend Edinburgh Scotland. Good Food, Good Drink, Good People. And you can hit most of the cool spots in a day.
either book a room at a hotel called the tulip inn next to Xcape, castleford for the night and go out around castelford/xcape or blackpool.but i must warn u its expensive!
Are you coming from the US en route to Europe, perhaps? Then it would make sense to visit a town near your airport of transfer, and if that is London, it makes good sense to explore that. Loads to see and do, huge choice of accommodation, easy transport options. Much as I love Norwich - my nearest city - by the time you got there from London it would be time to turn round and go back again!
Do you only have one day and night? If so, stick close to whatever city you are passing through. If that's London and you have only a single day consider one of the city bus tours. You'll get to see a lot, though you won't really "experience" it. If you love history then spending a day at the Tower of London or one of the museums is a good way to go. For a cheap day (London is way expensive) go to the area called "Embankment" and wander around--It's beautiful to walk along the water and you'll be close to Westminster Abbey, London Eye, Big Ben and House of Parliment. Also, lots of London Museums are free!
If you can go anywhere it really depends on what you like. The west coast of Scotland and the islands are beautiful but you can't do that in a day. I'm just guessing that maybe you are flying through and have a long layover, in which case you're probably passing through London. Have fun but come back for a longer visit sometime!
